Fast and efficient isolation of human EGFR-positive cells using EasySep&[trade]

Abstract Epidermal growth factor receptor (EGFR) is a cell surface normally expressed on most epithelial cells and some hematopoietic cells. Mutation overexpression of EGFR key driver many human cancers, including lung, breast, as well colorectal cancers. Circulating tumour (CTCs) can also express EGFR, where enrichment EGFR+ CTCs provide promising liquid biopsy prognostic values. EGFR-positive be difficult to isolate because they are present in tissue types at wide range frequencies. We have developed simple method address this challenge. A549 lung adenocarcinoma expressing were spiked into PBMCs defined frequencies model system. Starting with single-cell suspension 10% cells, the EGFR-expressing labeled an antibody complex that linked magnetic particles, then separated using EasySep™ magnet. Using method, enriched from 9.9 ± 3.3% 95.0 2.8% (mean SD; n = 14). Protocols been optimized for different sample sizes starting EasySep™-isolated viable proliferative, fold expansion Ki67 expression comparable untreated Human Positive Selection Kit enables easy isolation 15 minutes, facilitating cancer research.
